Differential and cytochemical analysis of blood cells in green frogs (Pelophylax ridibundus, Pallas, 1771)

U ovom radu koristila se puna krv zelene žabe Pelophylax ridibundus. Izračunala se brojnost eritrocita, leukocita te vrijednost hematokrita. Morfološka i kvantitativna analiza krvnih stanica provedena je bojanjem May-Grünwald Giemsom. Diferencijalnim citokemijskim tehnikama napravljena je citološka karakterizacija različitih tipova leukocita. Glikokonjugati i polisaharidi dokazani su tehnikama alcian blue (AB) i periodic acid Schiff (PAS). Stanični lipidi dokazani su tehnikom sudan black B (SBB). Enzimatsko-citokemijskim tehnikama dokazala se aktivnost alfa-naftil acetat esteraza (ANAE), alfa-naftil butirat esteraza (ANBE) te alkalne fosfataze (ALP). Brojnost i udio leukocita, hematokrit i morfologija stanica slični su ostalim žabama, dok je brojnost eritrocita veća. Bojanje ABom imaju jezgre svih stanica, citoplazma limfocita, monocita, dio neutrofila, trombocita i neke granule eozinofila. PASom se boje limfociti, granule eozinofila, neki monociti, neutrofili i trombociti. SBBom se boje leukociti, trombociti, slabije nezreli i zreli eritrociti. Aktivnost ANAE pokazuju monociti i neki limfociti. Aktivnost ANBE imaju monociti, neki trombociti i limfociti. ALP nije dala pozitivne rezultate.This thesis presents results attained from peripheral blood of the frog Pelophylax ridibundus. It contains hematocrit values, erythrocytes and leukocytes counts. The May-Grünwald Giemsom stain was used in morphological and quantitative blood cell analysis. Cytological characterization of leukocytes types used differential cytochemical stains. The alcian blue (AB) dye and the periodic acid Schiff method (PAS) stained glycoconjugates and polysaccharides. The sudan black B (SBB) dye stained cell lipids. Activities of alpha-naphthyl acetate esterase (ANAE), alpha-naphthyl butyrate esterase (ANBE) and alkaline phosphatase (ALP) were proved using enzyme-cytochemical reactions. Leukocyte count and percentage, hematocrit and cell morphology are similar to other frogs, unlike erythrocyte counts which are greater. AB stained nuclei of all cells, cytoplasm of lymphocytes, monocytes, some neutrophils, thrombocytes and some eosinophil granules. PAS stained lymphocytes, eosinophil granules, some monocytes, neutrophils and thrombocytes. All leukocytes, thrombocytes, young and mature erythrocytes stained with SBB. Monocytes and lymphocytes have active ANAE. Monocytes, some lymphocytes and thrombocytes have active ANBE. ALP activity was not recorded
