Cytotoxic and antioxidant effects of raspberry leaf extract on tumor cell lines HEp2 and SW 480

Učinci ekstrakta lista maline Rubus idaeus ispitivani su na dvjema staničnim linijama ljudskih tumorskih stanica, HEp2 (karcinom grkljana) i SW 480 (metastatske stanice karcinoma debelog crijeva). Cilj ovog rada bio je istražiti da li ekstrakt djeluje citotoksično na tumorske stanice i ako da, pri kojim koncentracijama ekstrakta, odnosno pri kojem vremenu tretmana. Ispitani su i uvjeti pri kojima ekstrakt djeluje antioksidacijski, odnosno kako prethodno izazvani oksidacijski stres utječe na razinu slobodnih radikala u stanici. Promatrane su promjene na razini ukupnog staničnog glutationa, koji je glavni stanični regulator razine oksidacijskog stresa, uslijed djelovanja ekstrakta lista maline. Vertikalnom elektroforezom utvrđeno je da produljenjem vremena tretmana dolazi do gubitka određenih citosolnih proteina. Ekstrakt lista maline pokazao je citotoksično djelovanje povećanjem vremena tretmana, a značajnije antioksidacijsko djelovanje pokazao je na HEp2 staničnu liniju. Niže koncentracije ekstrakta dovode do povećanja ukupnog glutationa u ispitivanim stanicama.The effects of raspberry leaf extract Rubus idaeus were examined in two human cancer cell lines, HEp2 (laryngeal carcinoma) and SW 480 (metastatic colorectal cancer cells). The aim of this study was to investigate whether the extract cytotoxic effect on tumor cells and if so, at which concentrations of the extract, or at what time of treatment. The conditions under which the extract expresses antioxidant activity were examined, and what is the influence of raspberry leaf extract on previously induced oxidative stress in the cells. The changes in the level of total cellular glutathione, which is an important regulator of levels of oxidative stress in the cell, were observed through the action of raspberry leaf extract. Vertical electrophoresis showed that extending treatment time leads to loss of certain cytosolic proteins. Raspberry leaf extract showed cytotoxic activity by prolonged the time of treatment, and it showed an significant antioxidant activity on the HEp2 cell line. Lower concentrations of the extract showed an increase of total glutathione in the tested cells
