Characterization of the biological effects of IL-22 and IL-22-BP and their immunological role during inflammatory reactions

Zytokine spielen eine wichtige Rolle während septischen Entzündungsreaktionen. IL-22 ist ein kürzlich entdecktes pro-inflammatorisch wirksames Zytokin. Die biologische Aktivität von IL-22 wird von IL-22-Bindeprotein (BP) moduliert, ein natürlicher Antagonist von IL-22. Hintergrund der Arbeit war die Rolle von IL-22 in einem septischen Peritonitismodell zu studieren. RT-PCR Analysen zeigten eine Induktion von IL-22 und dessen IL-22-Rezeptor in der Leber als auch in der Milz. Um die Wirkungsweise von IL-22 in der polymikrobiellen Sepsis näher zu beleuchten wurde Mäusen vor Sepsisinduktion rekombinates IL-22-BP verabreicht, um endogenes IL-22 zu blockieren. IL-22-BP wurde als Fc-gamma2a Fusionsprotein synthetisiert. Dies führte zu einer Verlängerung der biologischen Halbwertszeit in vivo. Die Vorbehandlung mit IL-22-BP/Fc-gamma2a 4h vor Sepsisinduktion führte zu einer Akkumulation von Neutrophilen und Monozyten/Makrophagen und einer verminderten bakteriellen Belastung der Bauchhöhle. Zudem führte die Blockade von IL-22 zu einer verminderten bakteriellen Belastung der Leber/Niere sowie einem geringeren Organschaden der Niere 12h nach Sepsisinduktion. Die hier gezeigten Ergebnisse lassen auf eine wichtige Rolle von IL-22 während der septischen Peritonitis schließen.Cytokines are causally involved in the natural course of inflammation during sepsis. IL-22 is a recently discovered pro-inflammatory cytokine. The biological activity of IL-22 is considered to be modulated by IL-22-binding protein (BP), which is regarded as a natural antagonist of IL-22. We studied the role of IL-22 in a model of polymicrobial peritonitis. RT-PCR analysis showed an induction of IL-22 and IL-22 receptor in both liver and spleen during the course of sepsis. To further analyse the role of IL-22 during septic peritonitis we pre-treated mice with recombinant IL-22-BP to block IL-22 action. IL-22-BP was generated as Fc-gamma2a-fusion-protein to prolong the half-life time in vivo. Pre-treatment of mice with IL-22-BP/Fc-gamma2a 4h before sepsis induction lead to enhanced accumulation of neutrophils and monocytes/macrophages and a reduced bacterial load in the peritoneal cavity. In addition, IL-22 blockade caused an enhanced bacterial clearance in liver and kidney and reduced kidney injury 12h after sepsis induction. These results imply an important role of IL-22 during septic peritonitis, contributing to bacterial spread and organ failure
