Cellular sources and role of Interleukin-22 Binding Protein (IL-22BP) in the control of homeostatic actions of IL-22 in the intestine

L'IL-22 est une cytokine produite les lymphocytes qui agit uniquement sur les cellules épithéliales des tissus de barrière comme la peau, les poumons et l’intestin. Au-delà de ses actions durant l'inflammation comme le renforcement de l’intégrité de la barrière épithéliale et l’induction des peptides antimicrobiens, l’IL-22 joue un rôle constitutif important dans le maintien de l'homéostasie intestinale. L’IL-22 possède un récepteur soluble inhibiteur appelé IL-22-binding protein (IL-22BP). Nous avons précédemment montré que l'IL- 22BP entrave les effets soit protecteurs soit délétères de l’IL-22 sur l’épithélium intestinal et cutané, respectivement durant l’inflammation. L’objectif de ce travail était de déterminer si l’IL- 22BP contrôlait aussi les actions homéostatiques de l’IL-22 au niveau intestinal. La première partie de ce travail a été consacrée à l’identification chez le rat des Innate lymphoid cells (ILCs) qui sont les principales cellules productrices d’IL-22 dans l’intestin. Nous avons ensuite revisité les sources cellulaires d’IL-22BP à l’aide d’un nouveau modèle animal et montré une expression restreinte aux phagocytes mononuclées. Enfin, nous avons démontré une augmentation des actions de l’IL-22 sur les cellules épithéliales intestinales en l’absence d’IL-22BP in vivo et ses conséquences sur la flore microbienne et les réponses T intestinales. Ces travaux indiquent que les actions de l’IL-22 sont en permanence finement contrôlées par l’IL-22BP qui participe ainsi à l’équilibre des interactions entre la flore et le système immunitaire.IL-22 is a cytokine produced by lymphocytes that acts only on the epithelial cells from barrier tissues such as the skin, lungs and intestine. Beyond its actions during inflammation such as enhancing the integrity of the epithelial barrier and the induction of antimicrobial peptides, IL-22 plays an important constitutive role in the maintenance of intestinal homeostasis. IL-22 has an inhibitory soluble receptor called IL-22-binding protein (IL- 22BP). We have previously shown that IL-22BP inhibits the protective or deleterious effects of IL-22 on the intestinal and cutaneous epithelium, respectively during inflammation. The aim of this work was to determine if IL-22BP also controls homeostatic actions of IL-22 in the intestine. The first part of this work was devoted to the identification in rats of Innate lymphoid cells (ILCs), which are the main cells producing IL-22 in the intestine. We then revisited the cellular sources of IL-22BP using a new animal model and showed that its expression was restricted to mononuclear phagocytes. Finally, we demonstrated an increase in the actions of IL-22 on intestinal epithelial cells in the absence of IL-22BP in vivo and its consequences on microbial flora and intestinal T cells responses. These studies indicate that the actions of IL-22 are permanently finely controlled by IL-22BP, which thus contributes to the balance of interactions between the flora and the immune system
