Heterotrophic freshwater bacterial groups and the biogeochemical cycling of phosphorus in lakes

Phosphor ist ein lebensnotwendiger Nährstoff, der die primär und bakterielle sekundäre Produktion in vielen ozeanischen Gebieten und den meisten Süßwasser-Ökosystemen bestimmt. Anorganisches Phosphat wird dabei als die bevorzugte Phosphor-Quelle für Mikroorganismen angesehen, kommt jedoch in aquatischen Systemen oft nur in sehr niedrigen Konzentrationen vor. Eine alternative Phosphor-Quelle sind gelöst organische Phosphorverbindungen. Heterotrophe Bakterien spielen eine zentrale Rolle im Nährstoff Kreislauf von aquatischen Ökosystemen. Sie mineralisieren totes organisches Material zu Biomasse und liefern so höheren Organismen Energie und Nahrung. Obwohl Phosphor eine entscheidende Rolle im Wachstumsverhalten von aquatischen Bakterien spielt, ist sehr wenig über die Identität von Süßwasser Bakterien in der Aufnahme von verschiedenen Phosphorverbindungen bekannt. Das Hauptziel meiner Dissertation ist es deshalb zu verstehen, welche ökologische Rolle den Süßwasser Bakterien in der Phosphor Aufnahme zukommt und welche Strategien unterschiedliche Bakteriengruppen verfolgen, um auch bei niedrigen Phosphorkonzentrationen kompetitiv zu sein. Dazu wurden die Bakteriengemeinschaften und die physikalisch-chemischen Bedingungen im Epilimnion und Hypolimnion von drei Bergseen und einem subarktischen See bestimmt, sowie eine umfangreiche Studie über die Aufnahme von anorganischen und organischen Phosphorverbindungen einzelner Bakteriengruppen durchgeführt. Die Ergebnisse der ersten Studie zeigen, dass Monomere mit Kohlenstoff und Phosphor und/oder Stickstoff eine wichtige Nährstoffquelle für das bakterielle Wachstum in oligotrophen Hochgebirgsseen darstellen (I). Ferner weisen sie darauf hin, dass die am häufigsten vorkommenden Bakterien Gruppen (AcI Aktinobakterien und R-BT-Cluster von Betaproteobakterien) auf bestimmte Substrate spezialisiert sind, was ihre Koexistenz in Süßwassern erklären könnte. In einer zweiten Studie wurde untersucht, ob die Aufnahme von anorganischen und organischen (ATP) Phosphorverbindungen einzelner Bakterien Gruppen ihrem Beitrag zur relativen Abundanz und Produktion entspricht (II). Es zeigt sich, dass AcI Aktinobakterien einen niedrigeren Phosphorbedarf als andere Süßwasser Bakterien haben, was ihre Verbreitung in Phosphor-limitierten Seen begünstigen könnte. Im Gegensatz scheint der R-BT-Cluster der Betaproteobakterien eine starke Homöostase unter Phosphor-reduzierten und -reichen Bedingungen zu halten, um unter Nährstoffeintrag sowie unter limitierenden Bedingungen dominieren zu können. In einer weiteren Studie (III) konnte gezeigt werden, dass Süßwasser-Bakterien neben ATP, auch Glukose-6-Phosphat und Glycerol-3-Phosphat in einem breiten Spektrum von natürlich vorkommenden Substratkonzentrationen nutzen können. Dabei unterscheiden sich einzelne Bakterien Gruppen aber deutlich in ihrer Fähigkeit diese Verbindungen aufzunehmen. Zum Beispiel, die AcI Aktinobakterien waren nur wenig in der Aufnahme von gelöst organischen Phosphorverbindungen beteiligt, während der R-BT-Cluster der Betaproteobakterien die Substrat-Aufnahme dominierte. Die Bedeutung von terrestrischen Eintrag für die Seen Produktivität und die Bakteriengemeinschaft wurde in einem Hochgebirgs- und Subarktischen See untersucht (IV). Die Resultate deuten darauf hin, dass klimabedingte Veränderung der Vegetation (Baumgrenze) und der Bodenzusammensetzung, die Bakteriengemeinschaft und deren Funktion im See erheblich beeinträchtigt. Dies ist insofern relevant, da Veränderungen im Boden in Bezug zur Phosphor- und Kohlenstoff-Bioverfügbarkeit, das Bakterienwachstum erheblich begünstigt und somit den Kohlenstoff und Phosphor Kreislauf des Sees stören könnte.Phosphorus is an essential nutrient for life that is known to limit both primary and bacterial secondary production in large oceanic areas and most freshwater ecosystems. Although inorganic phosphate is considered the preferred phosphorus source for microorganisms, utilization of labile dissolved organic phosphorus (DOP) is an alternative strategy to cope with low phosphate concentrations and to support biomass production. Heterotrophic bacteria play a central role in the biogeochemical cycle of elements in aquatic ecosystems. They mineralize dead organic material and thus, provide energy and nutrients to higher trophic levels. Despite the essential role of phosphorus in aquatic ecosystems and the common limitation of bacterial production by this nutrient, very little is known about the identity of freshwater bacteria involved in phosphorus uptake. The main aim of this thesis is to understand the role that heterotrophic freshwater bacteria play in the uptake of inorganic and organic phosphorus compounds, as well as the strategies that individual bacterial taxa use to cope with low phosphorus concentrations. Besides the description of the bacterial community composition and the prevailing physicochemical conditions in the epilimnion and hypolimnion of three mountain lakes and one subarctic lake, an extensive study on inorganic and organic phosphorus uptake patterns by individual bacterial taxa was done. The results of the first study demonstrate that monomers containing either C and N or C and P are an important resource to support bacterial growth in oligotrophic high mountain lakes (I). Further, they point to substrate partitioning among the most abundant bacterial taxa (AcI Actinobacteria and R-BT cluster of Betaproteobacteria) in freshwaters, helping to explain their coexistence by physiological niche separation. Whether the contribution of individual bacterial taxa to inorganic and organic (ATP) phosphorus uptake is proportional to their relative contribution to abundance and production was determined in the second study (II). AcI Actinobacteria appear to have lower phosphorus requirements than other freshwater bacteria, favouring their prevalence in phosphorus-limited lakes. In contrast, the R-BT cluster of Betaproteobacteria seems to keep strong homeostasis during phosphorus-deficient and phosphorus-sufficient conditions and thus, it does not only dominate at nutrient pulses, but it also persists during phosphorus-depleted conditions. In another study (III), we showed that ATP, Glucose-6-phosphate and Glycerol-3-phosphate were readily taken up by freshwater bacteria, proving that they can efficiently use DOP compounds at a wide range of naturally occurring substrate concentrations. However, when looking at individual bacterial taxa, they clearly differ in their ability to utilize these compounds. For instance, AcI Actinobacteria seem to be less involved in DOP acquisition, whereas the R-BT cluster of Betaproteobacteria dominates the uptake of low-molecular-weight DOP compounds. In the last study (IV), the importance of terrestrial carbon and nutrients for the productivity and for structuring the bacterial community of high altitude and latitude lakes was assessed. This study highlights that climate-induced changes in lake catchments affecting the availability of phosphorus and carbon in their soils, have a strong impact on bacterial community composition and production rates and thus, on the flux of carbon and phosphorus to higher trophic levels.by MSc.
