Caractérisation fonctionnelle de gènes impliqués dans la tolérance au stress métallique chez les champignons ectomycorhiziens par agrotransformation d'Hebeloma cylindrosporum

The purpose of my work was to develop a method of genetic transformation through Agrobacterium tumefaciens (agrotransformation) of the ectomycorrhizal fungus Hebeloma cylindrosporum to characterize the function of target genes. These genes were the zinc transport protein HcZnT1 of H. cylindrosporum, the thioredoxin PiTrx and the metallothionein PiMt1, from the fungus Paxillus involutus. The method of agrotransformation, which has a relatively easy setup compared to other methods of transformation, has proved to be effective and it was possible to obtain stable transformants with reproducible rates of transformation. The cloning of genes in the plasmide pFAT3 revealed to be more favourable to carry out the transformation than the other binary vectors tested (pBGgHg, pBINplus) which could not be used to clone the target genes. For each gene it was possible to harvest stable transformants harboring the transgene. The analysis by RT-PCR of transcripts revealed that transgenes were expressed. The phenotypical analysis revealed that the PiMt1 gene is able to increase the tolerance capacity of H. cylindrosporum to copper, but not to the other tested metals (Zn, Cd, Ni).L'objectif de mon travail de thèse a été de mettre au point une méthode de transformation génique par Agrobacterium tumefaciens (agrotransformation) du champignon ectomycorhizien Hebeloma cylindrosporum en vue de caractériser la fonction des gènes utilisés. Les gènes cibles sont le transporteur de zinc HcZnT1 d'H. cylindrosporum, la thiorédoxine PiTrx et la métallothionéine PiMt1, du champignon Paxillus involutus. La méthode de l'agrotransformation, dont la mise en oeuvre est relativement facile par rapport aux autres méthodes de transformation, s'est avérée efficace et il était possible d'obtenir des transformants stables avec des taux de transformation reproductibles. Le clonage des gènes dans le plasmide pFAT3 s'est montré plus favorable pour réaliser la transformation que les autres vecteurs binaires testés (pBGgHg, pBINplus) qui ne pouvaient pas être utilisés pour cloner les gènes. Pour chaque gène il était possible de recueillir des transformants stables contenant le transgène. L'analyse par RT-PCR des transcrits révèle que les transgènes sont effectivement exprimés. L'analyse phénotypique a fait apparaître que gène PiMt1 est capable d'accroître la capacité de tolérance d'H. cylindrosporum vis-à-vis du cuivre, mais non des autres métaux testés (Zn, Cd, Ni)
