Zymogram analysis of protease production in the presence of protease inhibitors.

<p>Analysis of the production of specific proteases was assessed using casein (upper panel) and gelatin gels (lower panel) in UAMS-1 (WT) and its UAMS-929 <i>sarA</i> mutant. Analysis was done after growth in biofilm medium in the absence of any inhibitor (Φ), the presence of the inhibitor cocktail (CT), or the presence of each inhibitor alone and in combination with each other. Inhibitor designations are the same as in the legend for <a href="http://www.plosone.org/article/info:doi/10.1371/journal.pone.0003361#pone-0003361-g010" target="_blank">Fig. 10</a>. The presumed identity of individual proteases is indicated to the right. In the case of SspA, this was confirmed by analysis of the corresponding <i>sarA/sspA</i> mutant (data not shown). In the case of ScpA and SspB, identification is presumptive based on their identity as cysteine proteases, inhibition of both by E-64, and the fact that ScpA has greater activity than SspB on casein while the opposite is true on gelatin (Dr. Jan Potempa, personal communication).
