Analysis of influenza virus NP-specific CD8 T-cell responses in the lungs of rAd/NP-immune mice.

<p>BALB/c mice were immunized intranasally or sublingually with 1×10⁷ PFU or 1×10⁸ PFU of rAd/NP. Control mice were immunized intranasally with rAd/Mock. Mice that received rAd/NP via sublingual route were boosted two weeks after the primary vaccination. Subsequently mice in all vaccination groups were challenged intranasally with 10 LD₅₀ of PR8 virus three weeks after their last immunization. (A) At day 5 post-challenge, lung cells were harvested from mice (n = 5) in each group and stained with K ^d/NP_147-155 tetramer, anti-CD8, and anti-CD44 antibody. The percentage of K ^d/NP_147-155 tetramer-positive, CD44-positive cells among total lung CD8-positive cell population was indicated in the upper right quadrant. (B) The average percentages of K^d/NP_147-155 tetramer-specific CD8 T cells in the lungs of five mice per group.
