PP1-γ silencing impairs NF-κB and MAPK signaling events downstream of TLR activation.

<p>A) HEK293T/TLR7/NF-κB-luc cells were reverse transfected with the indicated siRNAs. Forty-eight hours post-transfection, cells were left untreated, or stimulated with R848 (0.5 µM) or TNF-α (10 ng/mL) for 40 minutes, then fixed and stained. Nuclear translocation of p65 was evaluated by immunofluorescence (nuclei (red) = DAPI; p65 (green)). B–C) HEK293T/TLR7/NF-κB-luc cells were reverse transfected with the indicated siRNAs. Forty-eight hours later, cells were stimulated with R848 (10 µM) for the indicated time points, and whole cell lysates were collected and used for immunoblotting with the indicated antibodies.
