The Auxiliary-mediated Michael reaction : solid phase synthesis and control of stereochemistry

Der Aufbau quartärer Stereozentren gelingt mittels eines neuen Valinamid-Auxiliars in asymmetrischen Michael-Reaktionen mit bis zu 99% ee. In dieser Arbeit wird über die Anbindung des Auxiliars an einen polymeren Träger und dessen Verwendung in asymmetrischen Michael-Reaktionen berichtet. Bei unsymmetrischen beta-Diketonen als Michael-Donoren ist die Anbringung des Auxiliars an zwei Ketogruppen möglich. Gezielte Kontrolle der Regioselektivität der Auxiliaranbindung ermöglicht die stereoselektive Bildung beider Enantiomere eines Michael-Produkts mit lediglich dem natürlich vorkommenden Enantiomer des Auxiliars.The stereoselective construction of quaternary stereocenters can be achieved by means of a novel valine amide auxiliary with up to 99% ee. In this thesis the auxiliary is fixed onto a solid support and its use in asymmetric Michael reactions is reported. When using non-symmetric beta-Diketones as Michael donors, the auxiliary can be attached to the substrate at two different keto groups. Control of the regioselectivity of the enamine formation leads to the stereoselective construction of either enantiomer of the respective Michael product applying only the natural enantiomer of the chiral auxiliary
