Modification of Human Blood Hematocrit Level to Optimizing Streptococcus Pneumoniae Growth on the Human Blood Agar Media

Background: S. pneumoniae an important pathogen in humans because it can cause pneumoniae, sinusitis, otitis, bronchitis, bacteremia, meningitis. Sheep blood agar is the standard media for microbiological examination. Difficulties in supply of sheep blood in tropical countries is a constraint culturing S. pneumoniae, whereas the use of human blood that provision is not recommended for culture. This study aims to find an alternative to culture medium of human blood with hematocrit levels of medicationMethods: This study uses experimental design True post test only. Ninestrains of S. pneumoniae grown on media HuBA-St,SBA, and HuBA-Ht then incubated for 24 and 48 hours. Colony diameter, the diameter of hemolysis and colony characteristics are assessed in the incubation media to spike sputum.Results: After incubation 24 and 48 hours showed the colony diameter of S. pneumoniae in HuBA-Ht media is greater than HuBA-St and SBA (p = 0.000) in sputum spike suspension. Characteristics oftypical colonies of S. pneumoniae that in spike into the sputum in the HuBA-St and SBA to the observation of 24 and 48 hours of incubation is not significantly different 24 hours (p = 0.570) and 48 hours (p = 0.347)Conclusion: Media for the modification of human blood hematocrit as a viable alternative to the culture medium of S. pneumoniae in sputum spike can grow S. pneumoniae with characteristics maintained
