volume 8 Number 211980 Nucleic Acids Research Transfer RNA genes in the cap-oxil region of yeast mitochondrial DNA

A cytopiasmic "pet i te " (p~) clone of Saccharomyces cerevisiae has been Isolated and found through DNA sequencing to contain the genes for cysteine, h is t id ine, leudne, glutamine, lys ine, arginine, and g lydne tRNAs. This clone.designated DS502, has a tandemly repeated 3.5 kb segment of the wi ld type genome from 0.7 to 5.6 uni ts. Al l the tRNA genes are transcribed from the same strand of DNA in the direct ion ca_p _ to ox1l. The mitochondrial DNA segment of DS502 f i l l s a sequence gap that existed between the h is t id ine and lysine tRNAs. The new sequence data has made 1t possible to assign accu-rate map positions to a l l the tRNA genes in the cap-oxil span of the yeast mitochondrial genome. A detailed res t r ic t ion map of the region from 0 to 17 map units along with the locations of 16 tRNA genes have been determined. The secondary structures of the leucine and glutamine tRNAs have been dedu-ced from the i r gene sequences. The leudne tRNA exhibits 64 % sequence homol-ogy to an £. co l l leudne tRNA
