Bu durum, olguların yaklaşık %90’ında mayoz süresince maternal kromozom ayrılamamasından kaynaklanmaktadır. Kromozomal ayrılamama ve folat metabolizması arasındaki ilişki son yıllarda ilgi çekmeye başlamıştır.
