Braf v599e mutations incidence in nevi cases

BRAF, RAF gen ailesindendir ve 7. kromozomun uzun kolunda lokalizedir (7q34). BRAF geni MAPK sinyal yolunda görevli ve serin/treonin kinaz aktivitesi olan bir protein kodlar. BRAF geninde meydana gelen çeşitli mutasyonların MAPK sinyal yolunun aktivitesini artırarak, çekirdek içi transkripsiyon faktörlerinin aktivasyonu ile hücre çoğalmasına neden olduğu düşünülmektedir. Melanomlarda tanımlanmış olan BRAF ekzon 15, T1796A transversiyonu, 599 numaralı valin`in yerine glutamik asit`in kodlanmasına neden olur. Literatürde V599E mutasyonunun melanoma örneklerinde görülme sıklığının %90`ın üzerinde olduğu belirtilmektedir. Bu tez çalışmasının amacı, nevüs olgularında BRAF gen mutasyonu varlığının araştırılmasıdır. Pamukkale Üniversitesi Tıp Fakültesi, Patoloji Anabilim Dalında nevüs tanısı almış 30 olgu çalışma kapsamına alınmıştır. Parafin kesitlerden DNA izolasyonunu takiben, BRAF geninin 1796 nükleotid bölgesini içeren DNA parçası polimeraz zincir reaksiyonu ile çoğaltıldıktan sonra DNA dizi analizi ile mutasyon varlığı araştırılmıştır. Ayrıca aynı olgular Anti-ACTİVE® MAPK pAb Rabbit, (pTEpY) antikoru kullanılarak immunohistokimyasal yöntemle boyanarak olası MAPK sinyal yolu aktivasyonu araştırılmıştır. 30 olgunun 5 tanesinde (%16.6) T1796A transversiyonuna bağlı V599E mutasyonunun varlığı tespit edildi ve immunohistokimyasal analizde bu olgulardaki mutasyona bağlı MAPK sinyal yolu aktivasyonunun olmadığı gözlemlendi. Bu araştırmada elde dilen sonuçlar nevüslerde BRAF V599E mutasyonu sıklığının melanomlardaki kadar yüksek olmadığını göstermektedir. Ancak Türk hasta populasyonu yönünden kesin bir yargıya varılabilmesi için bu çalışmadaki örnek sayısının artırılması gerekmektedir.BRAF belongs to the family of RAF gene located at long arm of chromosome 7 (7q34). The BRAF gene encodes a protein that has function in MAPK signaling pathway with serin/threonin kinase activity. It has been thought that diverse mutations occurring in BRAF gene may activate MAPK signaling pathway which results in nuclear transcription factors activation. T1796A transversion of BRAF gene has been defined in melanomas that causes missense coding of glutamic acid rather than valin at aminoacid at position 599. It is suggested that incidence of the V599E mutation in the melanoma samples is nearly ninety percent (%90). The aim of this thesis study is to analyze incidence of BRAF T1796A gene mutation in nevi samples. Thirty nevi cases, diagnosed at the department of Pathology Pamukkale University Medical Faculty, were included in this study. After DNA isolation from the paraffin blocks, presence of BRAF V599E mutation is investigated by the amplification of DNA segment including 1796. nucleotide region with polymerase chain reaction and direct DNA sequencing. The same samples also stained with Anti-ACTİVE® MAPK pAb Rabbit, (pTEpY) antibody in order to analyze MAPK signaling pathway activation. The presence of V599E mutation depending on the T1796A transversion is determined in five of thirty cases (%16.6), and the immunohistochemistry does not support the MAPK signaling pathway activation depending on the mutation in these cases. Results of this thesis study suggest that the incidence of BRAF V599E mutation in nevi samples is not as high as described in melanomas. However, for a certain decision on the Turkish patient population, the number of samples included in this study should be significantly increased
