Transcription is the first step of gene expression. In eukaryotes, messenger RNA (mRNA) transcription is a highly regulated process. Transcription begins with the binding of a specific transcription factor on a DNA regulatory sequence. This enable the recruitment of co-activators, followed by general transcription factors (GTFs) and RNA polymerase II (Pol II) to form preinitiation complex (PIC). Mediator is a co-activator complex which is essential in this process. In yeast Saccharomyces cerevisiae, Mediator is composed of 25 subunits, among which 10 are essential for cell viability, organized into four distinct modules.
