Note:To investigate the mechanisms regulating axonemal motility, a panel of monoclonal antibodies (MoAb) against axonemal proteins from Chlamydomonas reinhardtii has been prepared. MoAb L2 and L3 each recognized a 110 kDa protein and were shown to cause a marked decrease in the percentage of motile demembranatedreactivated Chlamydomonas. In contrast, MoAb L6 recognized a 34 kDa protein and decreased flagellar beat frequency. Studies on Chlamydomonas mutants showed that the L2 and L3 antigens were located on the stem and head of the radial spokes, respectively, while the L6 antigen was localized directly on microtubules. Screening a cDNA expression library using MoAb L6 allowed the detection of a cDNA encoding for the L6 antigen. Our results...
