Thalamic contribution to social emotion

There is substantial evidence that disturbances in learning and memory are associated with a damaged mediodorsal (MD) thalamus. On account of its strong afferent input from the central nucleus of the amygdala, the MD thalamus has also been implicated in the expression of fear-related emotions. Furthermore, this nucleus communicates with the prefrontal cortex, namely the cingulate and orbitofrontal cortex. The paraventricular (PVT) thalamus similarly communicates with the amygdala and prefrontal cortex, more precisely the central amygdala and limbic cortices, suggesting it plays a role in negative emotional states. Due to these anatomical connections, we set out to explore the effect of MD and PVT thalamic lesions on positive social behavioural interactions, as well as negative emotions such as fear and anxiety. Groups of Long-Evan rats received bilateral excitotoxic lesions of the MD or PVT thalamus through N-methyl-D-aspartate (NMDA) injections, or received sham control surgery. We then tested the rats on a variety of tasks to measure social and non-social emotional behaviours. Non-social, negative aspects of emotions were quantified through the elevated plus maze and auditory fear conditioning. Social emotions were measured with an indirect and direct social interaction task. We found that, although these thalamic areas may contribute to anxiety on some level, there was no evidence to indicate that these thalamic nuclei contribute substantially to fear or social emotion. Ce noyau communique aussi avec le cortex préfrontal, c'est-à-dire le cortex cingulaire et orbitofrontal. D'autre part, le thalamus paraventriculaire (PVT) communique également avec l'amygdale et le cortex préfrontal, soit le noyau central de l'amygdale et les cortex limbiques. Des lésions à ce segment du thalamus mènent à des modifications de l'état d'éveil, du comportement de dépendance et des émotions. Étant donné ces relations anatomiques, nous avons entrepris d'explorer l'effet des lésions au MD et PVT sur les interactions sociales, et sur les émotions négatives comme la peur et l'anxiété. Des groupes de rats Long-Evan ont donc reçu des lésions excitotoxiques bilatérales au MD et PVT via des injections d'acide N-méthyl-D-aspartique. Les rats ont ensuite été évalués avec des tâches quantifiant le comportement émotionnel social et non social. Le comportement émotionnel négatif et non social a été mesuré grâce au labyrinthe en croix surélevé et à une tâche de conditionnement à la peur. Le comportement émotionnel positif et social a été mesuré à l'aide de tâches d'interactions sociales directes et indirectes. Malgré le fait que ces noyaux du thalamus contribuent possiblement à certains aspects d'anxiété, nous avons déterminé qu'il n'y avait aucune preuve indiquant que ces noyaux contribuent à la peur ou aux émotions sociales.
