Antibiotics have to permeate across or into the bacterial cell envelope in order to reach their target sites. The Gram-negative bacterial cell envelope however works as a formidable barrier, often limiting sufficient antibiotic drug levels. An in-depth understanding of anti-infective permeation processes into or across the envelope (structure-permeability relationships) would therefore be of considerable interest allowing for complementary information to established efficacy testing (structure-activity relationships), together facilitating a more rational and optimized drug design process.
