Little attention has been paid to a possible relationship between lysosomal function and stimulation of secretory processes in endocrine cells. The last few years it has become increasingly evident that the secretion of insulin from the pancreatic beta-cell is the result of a very complex cascade of events, the details of which are far from elucidated and indeed may include the participation of the lysosomal system. We report here, with a combined in vitro and in vivo approach, that selective inhibition of islet lysosomal glycogenolytic acid glucan-1,4-alpha-glucosidase activity by the long-acting 1-deoxynojirimycin derivative emiglitate induces a profound suppression of nutrient-induced insulin release. In islet homogenate emiglitate stron...
We have studied, by a combined in vitro and in vivo approach, the relation between the inhibitory ac...
Aminooxyacetate, an inhibitor of cytosolic transamination reactions, inhibited insulin release evoke...
Chronic exposure (24-72 hrs) of pancreatic islets to elevated glucose and fatty acid leads to glucol...
In previous in-vivo studies we have presented indirect evidence for the involvement of islet acid gl...
The mechanism of nutrient-evoked insulin release is clearly complex. One part of that mechanism is p...
Accumulated evidence links an important signal involved in glu-cose-stimulated insulin release to th...
Aims/hypothesis The mechanisms by which glucose regulates glucagon release are poorly understood. Th...
Altered secretion of insulin as well as glucagon has been implicated in the pathogenesis of Type 2 d...
Beta cells situated in the islet of Langerhans respond more vigorously to glucose than do dissociate...
1. Several recent in vitro studies have suggested that production of nitric oxide (NO) from the isle...
Iodoacetate inhibits glyceraldehyde-3-phosphate dehydrogenase activity in pancreatic islets and caus...
Glucose increases insulin secretion by raising cytoplasmic Ca(2+) ([Ca(2+)](i)) in beta-cells (trigg...
Although insulin secretion is usually regarded as a Ca2+-dependent mechanism, recent studies have su...
There is a substantial intracellular degradation of insulin in pancreatic islets. This may be a phys...
An increase in the production rate of reduced pyridine nucleotides is currently considered as a coup...
