Ischemic and traumatic brain damage are major causes of disability and death. While much effort has been spent to develop pharmacological treatments for these conditions, no neuroprotective drugs are in clinical use. Neuronal death following ischemia and trauma occurs in selected cell populations of the brain at various time points after the insult. It is believed that death of neurons are in part regulated through the expression of genes and proteins which regulate cellular viability. The tumor suppressor p53 has a central role in regulating the cell cycle and DNA repair processes following DNA damage and hypoxia. p53 can also initiate death-promoting mechanisms within cells.
