Modifying synaptogenesis and functional state of neural networks \(\textit {in vitro}\)

Die aktuelle Studie definiert die Auswirkungen von antipsychotischen Behandlungen auf der Ebene der lokalen Netzwerke Schaltungen und Funktion. Die Verarmung von Chondroitinsulfaten und Hyaluronsäure in reifen neuronalen Kulturen unterbrach die perineuronalen Netze und verringerte die Dichte sowohl der glutamatergen als auch der GABAergischen Synapsen.The current study defined the effects of antipsychotic treatments at the level of local networks circuitry and function. Two antipsychotic drugs were studied: Haloperidol (first generation neuroleptic) and Olanzapine (second generation antipsychotic). Both treatments adjusted the excitation and inhibition balance within local neural circuits, by differentially modifying synaptogenesis and spontaneous activity of excitatory and inhibitory neurons populations. Haloperidol altered the wiring and activity of local networks in the inhibition favoring mode, while Olanzapine induced network synchronization, exhibiting a balanced effect on the excitatory and inhibitory neuronal populations. Furthermore, synapse restricting functions of extracellular matrix were re-addressed in this study. The depletion of chondroitin sulfates and hyaluronic acid in mature neuronal cultures disrupted perineuronal nets and decreased the density of both glutamatergic and GABAergic synapses
