P63 is a transcription factor of the p53 family with key roles in embryonic development, stem cells and cancer. P63-deficient mice fail to form stratified and glandular epithelia, including the prostate, demonstrating a critical function for this protein in prostate development. The P63 gene encodes two different isoform groups, TA and N, which can act as tumor suppressors or oncogenes respectively in different tumors. In the prostate gland, Np63 is the main isoform expressed specifically in basal stem cells. There is evidence that p63+ basal stem cells are the cells-of-origin in prostate cancer.
