POTENSI TUNIKATA Rhopalaea sp SEBAGAI SUMBER INOKULUM BAKTERI ENDOSIMBION PENGHASIL ANTIBAKTERI; 1. KARAKTERISASI ISOLAT

The research on potency of Tunicate Rhopalaea sp as a source of inoculum of endosymbiotic bacteria that produces antibacteria has been conducted. This research was intended to know the potential of tunicate as a source of bacteria???s inoculum and the character of symbiotic isolates in tunicate Rhopalaea sp. This paper is first to discuss characterization of the endosymbiont bacteria of Rhopalaea sp. The isolation of endosymbiont bacteria is done by using Marine Agar (MA) medium. The characterization of symbiotic bacteria included colony???s morphology and gram staining. The biochemical essay including : the MR-VP, the motility, the catalase, the TSIA, the citrate, and the urea test. Test of inhibition zone was done towards Salmonella thypi and Staphylococcus aureus. We isolated 12 isolates (Code Asc1 ??? Asc12) of symbiont in tunicate Rhopalaea sp. The result in gram staining shown that 11 isolates were baccil, 10 isolates were gram-positive and 1 of those 11 isolates was gram-negative and 1 is gram-negative coccus. In the biochemical test reveals that 12 isolates were negative in the MR test, the indol as the citrate test, but they are positive in both the VP and the urea test, and can ferment glucose in TSIA medium. The catalase and the motility test show a various result. Asc4 is categorized as genus Alteromonas and the others were assumed to belongs to a genus Bacillus. All isolates were able to inhibit the growth of Salmonella thypi and Staphylococcus aureus bacteria
