Lipase (EC 3.1.1.3) is one of a class of hydrolase enzymes that can hydrolyze triglycerides into fatty acids and glycerol. This study aims to explore the lipolytic enzyme-producing microbes from hot springs Lemo Susu, Pinrang South Sulawesi, by identify microbes generated and determine the optimum time conditions of enzyme production. Production is done by growing microbes on the substrate medium olive oil. Produced lipase activity was tested by titrimetric method. The results obtained showed that the microbial isolates from hot springs Lemo Susu Pinrang, has identified as Bacillus sp. LS3B and Bacillus sp. LS4B.
