Signaling through the Tie2 receptor in endothelial cells has been shown to play an important role in normal and pathological vascular development. Therapeutic studies using a decoy Tie2 receptor suggest blocking of ligand binding to Tie2 inhibits tumor angiogenesis. The mechanism by which Tie2 inhibition affects tumor growth remains unclear. We generated an inducible soluble Tie2 receptor (Tie2Ex) in K1735 murine melanoma tumor cells to study the effects of inhibiting Tie2 signaling on tumor vasculature. Tie2Ex induction decreased tumor endothelial cell (EC) AKT activation but did not affect ERK activation.
