Advanced water soluble BODIPY dyes : synthesis and application

[...]Nous nous sommes intéressés au développement de nouvelles méthodes permettant de synthétiser des fluorophores hydrosolubles émettant dans le visible ou le proche infrarouge pour des applications en marquage biologique. Dans un premier temps nous avons effectué une étude méthodologique sur les modifications des boradiazaindacènes (BODIPYs) pour les rendre hydrosolubles et ceci tout en gardant leurs propriétés optiques. Dans un deuxième temps, en utilisant ces nouvelles méthodes nous avons développé une série de BODIPYs hydrosolubles émettant dans le rouge pour le marquage biologique. Le projet s’est ensuite orienté vers la synthèse de systèmes « push-pull » à partir d’un BODIPY. Les propriétés optiques et électrochimiques des nouveaux chromophores ont été étudiées.[...][...]The main theme of this thesis is to develop a set of water-soluble fluorophores as molecular probes for biological labeling applications. Despite the existing strategies which allow improving the water solubility of certain BODIPY dyes,1-3 some of these require multi-step synthesis or hash experimental conditions (e.g. using chlorosulfonic acid). Furthermore, most of these protocols can hardly be applied to more conjugated BODIPY dyes emitting in the red region of the electromagnetic spectrum in the 600 to 700 nm range. The main theme of this thesis is to develop a general strategy without the inconveniences mentioned above. This work was based on the functionalization of 4,4-difluoro-4-bora-3a,4a-diaza-s-indacene (BODIPY) as fluorescent framework, in order to introduce water solubility and functional groups to: (i) tune the color; (ii) to graft the probes to bio-molecules; (iii) to increase the dipole moment in the ground and excited states.[...
