Studies on imbalance localization of phospholipids in the cellular membrane

2009-2011年度科学研究費助成事業(科学研究費補助金(基盤研究(C)))研究成果報告書 課題番号：21580099 研究代表者：玉置尚徳 (鹿児島大学農学部准教授)本研究においては、細胞膜リン脂質に含まれる二重結合の不均衡な分布と我々が見出したアシル転移酵素との接点を解明した。酵母において見出した２種類のアシル転移酵素Lpt1とSlc1の高発現株より酵素を精製あるいは部分精製し基質特異性を調べた。また、それぞれの遺伝子破壊株をより抽出した脂質をLC-MS/MSを用いて解析した。その結果、両酵素は極性基の選択性は異なるもののリゾリン脂質に優先的に２重結合を導入することが明らかとなった。In this study, we clarify the relation between imbalance localization of double-bond in the phospholipids and newly found lyso-phospholipid acyltransferases. Two acyltransferases Lpt1 and Slc1 were purified (or partially purified) and substrate specificities were examined. Also total lipids were extracted from lpt1 or slc1 gene disrupted strain and subjected to LC-MS/MS analysis. From these experiments, Two enzymes mainly catalyze introduction of acyl-residue containing double-bond, although substrate preference were different
