Three affinity matrices for adsorption of endooxin from aqueous solution were studied by covalently bonding polymyxin B varying hydrocarbon lengths to Sepharose CL-4B. The content of ligand and adsorption efficiency increased with increasing length of spacers. The effect of salt, pH, temperature and flow rates on endotoxin adsorption for affinity matrix 3 was investigated. It showed that in the range of NaCl concentration of 0.05-0.5 mol/L, pH 6-10, temperature 25-55 degrees C and flow rate of 0.20-0.80 mL/min, the adsorption efficiency was over 90%
