An extraction method of triterpenoids in the fruiting bodies of ganoderma lucidum using supercritical fluid (SF) was developed. The effects of pressure, temperature and time were investigated with respect to extraction yield. The optimized parameters were as follows: extraction pressure 15 MPa, extraction temperature 35 degrees C, extraction time 120 min, flow rate of CO2 1 mL/min, and the temperature of backpressure regulator 50 degrees C. In addition, a gradient elution method of high performance liquid chromatography was developed for the analysis of extracted triterpenoids.
