The determination of the accurate value of hemoglobin concentration is a prerequisite in the diagnosis of anemia. Wong (1928) introduced an original method to determine hemoglobin iron which was modified in the present studies due to its lack in accurately determining the value of hemoglobin iron. The modified method is as follows. The iron of the hemoglobin molecule was detached completely by sulfuric acid and potassium permanganate without removing protein: Potassium persulfate and potassium rhodanatum were added and the quantity of iron was measured against the color of a standard solution of known iron content.
