One hundred and thirtyone faecal samples from cattle clinically suspected of having paratuberculosis were processed using the sedimentation culture method (SM) and the centrifugation culture method (CM) before inoculation into Herrold's egg yolk medium containing mycobactin J (M1), control medium as M1 with no mycobactin J (M2), medium as M1 plus miconazole (M3) and control medium for M3 which did not contain mycobactin J (M4). On M1 media the isolation rates were 40.4% for SM and 45.8% for CM, on M3 the isolation rates were 35.8% and 41.2%, respectively.
