Previous studies have shown that the iron-sulfur cluster is not necessary for catalysis, but the motif is highly conserved from bacteria to humans in DNA repair enzyme homologues, specifically in base excision repair enzymes like Endonculease III and MutY. Ntg1p and Ntg2p are the two Endonuclease III homologues and base excision repair enzymes in Saccharomyces cerevisiae. Both proteins contain the highly conserved helix-hairpin-helix motif, whereas only Ntg2p contains the iron-sulfur cluster of the Endonuclease III family.
