UnrestrictedLymphoid-specific protein tyrosine phosphatase (LYP) has been found to be one of the most potent inhibitors of T cell subpopulation activation. A gain-of-function single nucleotide polymorphism (SNP) C1858T has been shown to be involved in several autoimmune diseases. We compared the enzymatic activity of LYP and Pro, Glu, Ser, Thr-rich protein tyrosine phosphatase (PEST). In vitro biochemical studies showed that LYP could differentially hydrolyze several phosphatase substrates. We screened a small molecule library for potential LYP inhibitors. In addition, our siRNA studies confirmed that T-cell receptor signaling was enhanced once LYP expression got repressed
