Bioactivity Directed Isolation Of Secondary Metabolites Of Salvia Adenophylla And Salvia Verticillata Subsp. Amasiaca Plants And Investigation Of Their Antioxidant And Anticholinesterase Activities Including Semi-synthetic Derivatives

Tez (Yüksek Lisans) -- İstanbul Teknik Üniversitesi, Fen Bilimleri Enstitüsü, 2010Thesis (M.Sc.) -- İstanbul Technical University, Institute of Science and Technology, 2010Bu tez çalışmasında, ilk aşamada Salvia adenophylla Hedge & Hub.-Mor. ve Salvia verticillata subsp. amasiaca bitkilerinin sırasıyla aseton ve metanol ekstreleri hazırlanarak toplam fenolik ve toplam flavonoit miktarları tayin edildi ve antioksidan aktiviteleri lipid peroksidasyonu inhibisyonu ve DPPH serbest radikal giderimi aktivitesi yöntemleriyle, antikolinesteraz aktivitesi ise Ellman yöntemi kullanılarak belirlendi. Aktif ekstrelerin başlıca sekonder metabolitleri izole edilerek saflaştırıldı, yapıları çeşitli spektroskopik (NMR, kütle, IR ve UV) yöntemlerle belirlendi. S. verticillata subsp. amasiaca bitkisinden terpenik yapıdaki ferruginol, horminon, 7 α-asetilhorminon, oleanolik asit ve ursolik asit, 3-asetil- β-amirin, steroid yapıdaki β-sitosterol ve stigmasterol ile flavon yapısındaki salvigenin ve 6-hidroksi-apigenin-7,4’-dimetileter elde edildi. S. adenophylla bitkisinden elde edilen saf maddelerin ikisi diğer bitkiden de elde edilen oleanolik ve ursolik asit, diğerleri ise triterpenik yapıdaki α-amirin ve 3- β-hidroksi-urs-11-en-28,13- β-olid ve basit bir şekerli fenolik bileşik olan asetofenon-4-O- β-glukopiranozit olarak belirlendi. İkinci aşamada ise, bu iki Salvia türünden izole edilen triterpenik yapıdaki oleanolik asit den hareketle altı farklı türevi 3-asetil-oleanolik asit, 3-okso-oleanolik asit, 3-oksim-olean-12-en-28-oik asit, 3-asetil-oleanoil-28-klorür, 3-asetil-oleanoil-28-morfolid, oleanolik asit 3-metil eter hazırlandı ve yapıları spektroskopik analizlerle belirlendi ve saf bileşiklerin antioksidan ve antikolinesteraz aktiviteleri incelendi.In this study, in the first step, Salvia adenophylla and Salvia verticillata ssp. amasiaca plants will be exhausted by aceton and methanol. Total phenolic and flavonoid contents of each extract were determined. The antioxidant activity of the extracts was established by using lipid peroxidation inhibition, and DPPH free radical scavenging methods. Extracts were also screened for the anticholinesterase activity by Ellman method. The six compounds were isolated from extract of S. verticillata ssp. amasiaca. Their structures were identified ferruginol, horminone, 7 α-acetylhorminone, oleanolic acid and ursolic acid, 3-acetyl- β-amyrin, steroids β-sitosterol and stigmasterol, salvigenin and 6-hydroxy-apigenin-7,4’-dimethyl ether. Extract of S. adenophylla were identified to be oleanolic acid and ursolic acid which were also isolated from the other plant and the other three compounds were elucidated α-amyrin and 3- β-hydroxy-urs-11-en-28,13- β-olide, and Acetophenone-4-O- β-glucopyranozide. In the second step, it is aimed to prepare by semi-synthetically more active derivatives of triterpene oleanolic acid, oleanolic acid 3-acetyl-oleanolic acid, 3-oxo-oleanolic acid, 3-oxime-olean-12-en-oic-acid, 3-acetyl-oleanoyl-28-chloride, 3-acetyl-oleanoyl-28-morpholide, oleanolic acid-3-metil ether were prepared, and their structures were determined and their bioactivities were also investigated.Yüksek LisansM.Sc
