Determination Of Total Antioxidant Activities And Phenolic Compounds In Green And Black Teas Produced By Microwave Technology

Bu çalışmada; yeni bir teknoloji olan mikrodalga tekniği kullanılarak yeşil ve siyah çaylar elde edilmiştir. Ardından yeşil ve siyah çaylarda toplam antioksidan aktivitesi, fenolik madde ve duyusal analizler gerçekleştirilmiştir. Çay örneklerinin toplam antioksidan aktivitesi tayininde DPPH serbest radikal yakalama metodu kullanılmıştır. Toplam fenolik madde ve teaflavin içerikleri ise spektrofotometrik yöntemlerle belirlenmiştir. Siyah çay örneklerinin toplam fenolik madde ve antioksidan aktivite değerleri; sırasıyla 56-119 mg/g gallik asit eşdeğeri ve 0,43-0,95 mg/g troloks eşdeğeri arasında bulunmuştur. Yeşil çaylarda bu değerler; 93,18-130,16 mg/g gallik asit eşdeğeri ve 0,6839-1,4522 mg/g troloks eşdeğeri arasında değişmektedir. Yeşil çaylarda bulunan kateşinlerin HPLC analizleri gerçekleştirilmiştir. EGCG değeri 3,1-10,7 mg/100mg arasında, EGC değeri 2,4-5,1 mg/100mg arasında, ECG değeri 0,9-3 mg/100mg arasında ve EC değeri 0.6-1,5 mg/100mg arasında değişmektedir. Taze çay yaprakları ve açılmamış tomurcuktan mikrodalga tekniğiyle üretilen yeşil çayların daha yüksek kateşin ve antioksidan aktivitesine sahip olduğu görülmüştür. Tüm analizler için sezonun etkisi önemli bulunmuştur. Teaflavin analizleri Assam ve mikrodalgada üretilen siyah çayların daha yüksek teaflavin içeriğine sahip olduğunu göstermiştir. Yeşil ve siyah çay örneklerinin duyusal analizi; mikrodalga ile üretilen çayların diğer Türk tipi çaylardan daha lezzetli olduğunu göstermiştir. Mikrodalga teknolojisinin çay üretiminde alternatif bir yol olarak kullanılabileceği sonucuna varılmıştır.Teas represent a rich source of flavonoids which act as as antioxidant. In this study; a new technology, microwave, were used to produce green and black teas. The antioxidant potential, phenolic contents and sensory qualities of green and black teas were analysed. DPPH free radical scavenging activity was used to determine total antioxidant potential of tea samples. Total phenolic contents and theaflavin contents of teas were analysed by spectrophotomeric methods. The phenolic compounds and antioxidant activities of black teas were found between 56-119 mg/g gallic acid equivalent, 0,43-0,95 mg/g trolox equivalent respectively. Those for green teas were ranged from 93,18-130,16 mg/g gallic acid equivalent and 0,6839-1,4522 mg/g trolox equivalent respectively. Green teas were evaluated for individual tea phenolics, catechins using HPLC method. EGCG ranged from 3,1 to 10,7 mg/100mg, EGC from 2,4 to 5,1 mg/100mg, ECG from 0,9-3 mg/100mg, and EC ranged from 0.6 to 1,5 mg/100mg. The results indicated that microwave processed green teas with young leaves and an unopened bud have higher contents of catechins and higher antioxidant activities than other comman teas. Seosonal variation effects were found to be important for all analysis. Theaflavin analysis showed that microwave processed and Assam black teas contained higher teaflavin contents than others. The sensory analysis of green and black tea samples showed that microwave processed teas had more flavour than other teas. It could be concluded that using microwave technology can be used as an alternative process for tea manufacture.Yüksek LisansM.Sc
