Effect Of Different Cooking Methods On Phenolic Compounds And Antioxidant Activity Of Black Rice

Ancak bu yüksek içerik pişirme işleminden etkilenmekte ve antioksidan kapasitesinde azalmalar olmaktadır. Pişirme işlemi sonucunda bile siyah pirincin benzerlerine nazaran yüksek antioksidan içeriğine sahip olduğu görülmektedir. Bu çalışmada siyah pirincin fenolik içeriği ve antioksidan kapasitesi incelenmiş ve farklı pişirme yöntemlerinin fenolik içeriğini ve antioksidan kapasitesini nasıl etkilediği araştırılmıştır. Bu amaç doğrultusunda Türkiye’de satışa sunulan tek bir markanın siyah pirinci ile çalışılmıştır. Pirinç pişiricide (R), elektrikli düdüklü tencerede (E), tüplü ocak üstündeki düdüklü tencerede (D) ve tüplü ocak üstündeki teflon tencerede (N) olmak üzere 4 farklı ev tipi pişirme yöntemi kullanılmıştır. Siyah pirinç örneklerinin fenolik madde içerikleri ve antioksidan aktiviteleri çeşitli metotlar ile tespit edildikten sonra fenolik madde profilleri HPLC metodu ile belirlenmiştir. Çalışmada kullanılan siyah pirinç örneklerinin toplam fenolik madde, toplam flavonoid, toplam antosiyanin ve toplam antioksidan aktivite tayinleri için uygulanan metotlarda kullanılmak üzere ekstraktlar hazırlanmıştır. Uygun çözgen sisteminin hazırlanmasında literatürdeki çalışmalar esas alınarak ekstraksiyon için %1 HCl ile asitlendirilmiş metanol kullanılmıştır.In this study, phenolic content and antioxidant capacity of black rice and how phenolic content and antioxidant capacity is affected by different cooking methods were searched. For this purpose only brand for black rice which go on sale in Turkey and four different type of cooking methods such as rice cooker (R), electric pressure cooker (E), pressure cooker on a gas range (D) and teflon pot on a gas range (N) were used. First phenolic content and antioxidant activity of black rice samples, then phenolic profile by HPLC were determined. Phenolic extracts were prepared to use in total phenolic content, total flavonoid, total anthocyanin and total antioxidant activity assays. Based on the studies in literature, the suitable solvent system for the extraction of black rice samples was contained methanol which acidified with 1% HCl. At the end of this study, it is seen that black rice has a high amounts of polyphenol content. Nevertheless, this high amounts of polyphenol content were affected by cooking and antioxidant activity of samples were decreased. Even after cooking black rice still has high antioxidant content compared to similar foods.Yüksek LisansM.Sc
