This thesis tested the hypothesis that the synergistic effect of cisplatin in combination with radiation was due to cisplatin inhibition of repair of radiation-induced DNA damage. Two model cell systems were used. DNA repair inhibition caused by cisplatin and the fundamental mechanism of DNA double strand break repair were studied in human fibroblast cells. The effect of cisplatin on stress response and the induction of radiation resistance was studied in yeast. It was shown that normal human fibroblasts cells (AG1522), irradiated in the presence or absence of cisplatin adducts have biphasic DNA repair curves with a fast and slow component.
