Antimutagen of aloe plants

記事区分：原著[Author abstract]The antimutagenicity of substances from a species of Aloe was investigated, and an antimutagen was isolated. Methanol extracts from dried leaves of A. arborescens inhibited frameshift mutation caused by 3-amino-l-methyl-5H-pyrido [4, 3b] indole in Salmonella typhimurium TA98. An antimutagen isolated from the methanol extracts was identified as an anthraquinone, aloe-emodin. Aloe-emodin inhibited frameshift mutation at concentrations from 10^<-4> to 10^<-3>. Barbaloin, the monoglucoside of aloe-emodin, was inactive. Fresh plants of A. arborescens contained 1.17 μg/g aloe-emodin (wet weight). This antimutagen was detected in five of six other Aloe species investigated by high-pressure liquid chromatography. These results suggest that A. arborescens and other Aloe species might yield substances useful for the prevention of some kinds of cancer.[著者抄録]キダチアロエの抗突然変異原性について調べ,その活性成分の検索を行った。キダチアロエ葉のメタノール抽出物に強い突然変異抑制活性が認められた。この活性成分は石油エーテル相に移行し,続いて行ったシリカゲルカラムクロマトグラフィーによる純化の結果,活性成分としてエモジン型アントラキノンのアロエエモジンを単離,同定した。本成分は0.1 mM/plateの濃度で60.3%,1.0mM/plateの濃度で86.3%の突然変異原抑制率を示した。これに対してaloe-emodinのC-配糖体であるbarbaloinには全く抑制活性がみられなかった。従って,barbaloinは配糖体のままでは抑制活性を示さず非糖体の型になってはじめて抑制活性を示すものと思われる。またHPLCによる各種アロエの分析の結果,キダチアロエ以外にA. ferox, A. vera, A. eru, A. compactaにもaloe-emodinとその配糖体barbaloinが検出されたことよりこれらのアロエも同様の抑制活性を有すると推察される
