La vitamine B6 est d'une importance primordiale dans tous les organismes et agit, sous la forme de phosphate de pyridoxal (PPL), comme cofacteur pour de nombreuses enzymes métaboliques. La principale voie menant à la biosynthèse de la vitamine B6 fut récemment étudiée et implique deux enzymes, PDX1 et PDX2 pour la production directe de PPL. La régulation de cette machinerie complexe fut étudiée dans cette thèse. Nous avons montré que la partie C-terminale de PDX1, impliquée dans la liaison à une sous-unité voisine, est coopérative avec l'un des substrats de PDX1, ainsi qu'impliquée dans la catalyse du PPL.
