Laminins, major components of basement membrane are glycosylated heterodimers composed of three chains α, β and γ. Laminin α1 chain (LMα1), specific to LM111 trimer is essential in development of retinal inner limiting membrane. Loss of function and mutation of LMα1 negatively impact physiological angiogenesis, whereas its overexpression promotes colon cancer incidence, tumor cell proliferation, blood vessel maturation and fibroblast expansion. LMα1 stimulates VEGFA, Notch pathway components Dll4 and its targets Hey1 and Hey2 in tumor stroma.
