Identification of enterotoxigenic properties and antibiotic sensitivity of Staphylococcus aureus isolated from some foods

Micrococcaceae familyasında yer alan Staphylococus cinsinin üyesi olan Staphylococcus aureus insanlarda gıda zehirlenmelerinde önemli mikroorganizmalardan biridir. Gıda işletmeleri ile kurumlara ait büyük mutfaklarda hijyen eksikliğinin göstergesi olarak kabul edilir.Materyal olarak; 31 adet şarküteri ürünü, 10 adet dondurulmuş hazır köfte, 5 adet dondurulmuş et döner, 30 adet hazır yemek, 25 adet yaş pasta, 45 adet peynir, 28 adet dondurma ve 16 adet sütlü tatlı olmak üzere toplam 190 örnek kullanılmıştır.S.aureus analizi için Baird Parker (Oxoid CM0275) Agar kullanılmıştır. Sayısı 10³ kob/g'ın üzerinde olan örneklerin enterotoksin analizi, VİDAS Staph enterotoksin II (SET2; Biomerieux. REF 30 705 enzim bağlantılı floresans tekniği kullanılarak yapılmıştır. Ayrıca toksin varlığı saptanan gıdalardan izole edilen S. aureus suşunun vancomycin ve cefsulodin'e karşı duyarlılığı incelenmiştir.S. aureus açısından incelenen 190 adet gıda örneğinin 47'si (%24,7) pozitif olarak saptanmıştır. Düzeyi 1,0x103 kob/g'dan fazla olan 11 adet (%5,78) örnek enterotoksin içeriği açısından incelenmiş ve sadece 6'sında pozitif sonuç elde edilmiştir. İzole edilen 6 farklı S. aureus suşunun antibiyotik duyarlılık testi sonucunda da cefsulodin'e karşı duyarlılığın, vancomycin'e karşı olan duyarlılıktan daha fazla olduğu saptanmıştır.Staphylococcus aureus, which is the member of Staphylococcus geneus in the family of micrococcaceae, is the important causitive agent in food poisoning. Occurance of S. aureus in foods is an indicator of deficieny of hygen in food processing factory and big kitchens.Total 190 samples, which were 31 delicatessen product, 10 frozen ready meatballs, 5 roasted rolling meat, 30 ready to eat food, 25 cake with cream, 45 cheese, 28 ice cream and 16 desert with milk, were used.Baird parker agar (Oxoid CM0275) was used for S. aureus analysis. Samples, which has the S. aureus higher than 10³ cfu/g, were studied for enterotoxins analysis with VİDAS Staph enterotoxin II (SET2; Biomerieux. REF 30 705) enzyme linked fluorescence analysis technique (AOAC 070404/2007:06). In addition; the sensitivity of S. aureus, which was isolated from enterotoxin positive foods, was stated against to vancomycin and cefsulodin.Finally, 47 (24,7%) of 190 samples were found as S. aureus positive. 11 Samples (5,78%) has S. aureus higher than 1,0x103 cfu/g,was analysed for enterotoxins and only 6 of them were found positive.
