Coupling Scanning Transmission Electron Microscopy (STEM) with Electron Energy Loss Spectroscopy (EELS) enables to study the chemical structure and the physico-chemical properties of nanomaterials. However, under usual electron doses, the electron beam irreversibly damages radiation-sensitive materials, such as organic-inorganic, organic and biological components. This thesis provides novel methodologies for the damage-free analysis of such systems at a spatial resolution better than 10 nm. These methodologies were applied to the individual characterisation of Metal-Organic Framework (MOF) drug nanocarriers, with the aim of providing a better understanding of their mechanisms of biodegradation in physiological media and their drug loading. ...
