Dybjer et al. report that polygenic risk of type 2 diabetes is associated with dementia risk, in particular vascular dementia. Associations were strongest for non-carriers of the gene APOE epsilon 4. However, two-sample Mendelian randomization analyses did not support a causal relationship between type 2 diabetes and vascular dementia.
