The Hippo signal transduction pathway plays critical roles in regulating development, organ size, and cancer progression, and is broadly conserved across metazoans. Mammalian Sterile 20-like Kinase 12 (MST1/2) are mammalian homologues of the Drosophila Hippo kinase, and along with downstream Large Tumor Suppressor Kinase 12 (LATS1/2), constitute the core kinases of the Hippo pathway. How MST1/2 are regulated, and in particular, what mechanisms connect them to upstream signaling components is a key question in the field.
