The MRN complex (MRE11-RAD50-NBS1) plays a central role in response to DNA double-strand breaks. We identified a patient with developmental defects and bone marrow failure carrying biallelic RAD50 mutations. One of the mutations creates a null allele while the other (noted RAD50E1035del) leads to the expression of a protein that has lost a single residue in one of the repeated heptad of the coiled-coil domain of RAD50. We show that RAD50E1035del represents a separation-of-functions mutation that impairs the DNA repair, the DNA replication and the resection warranted by the MRN complex whithout affecting its ability to activate ATM.
