Objective: To optimize the therapeutic dose and time window of picrosede II in cerebral ischemic injury in rats by orthogonal test. Methods The forebrain ischemia model was established by a bilateral common carotid artery occlusion (BCCAO) Method: Rats were randomly grouped according to orthogonal experimental design and treated by injecting picroside II intraperitonenally at different ischemic times with different doses. The concentration of lipid perioxide malondialdehyde (MDA) in serum and brain was determined by thiobarbituric acid assay. The activity of superoxide dismutase (SOD) in these samples was determined by xanthine oxidase assay.
