Production, purification et caractérisation partielle des antibiotiques sécrétés par la souche Saccharothrix tamanrassentensis DSM 45947 d'origine saharienne

The strain Saccharotrix tamanrassetensis DSM 45947, designated SA198, is a new actinobacteria species of Saharan origin, which in previous study was found to produce in the ISP2 complex medium three new antibiotic compounds with antifungal and antibacterial activities. The present study aims to explore the antimicrobial potential of the strain, to improve the antimicrobial activity production, to purify and characterize the produced molecules. The strain showed an interesting antagonist activity, in particular against pathogenic germs. The antimicrobial potential of the strain was examined by PCR for the presence of some genes responsible for antibiotic biosynthesis. The non-ribosomal peptide synthetase (NRPS) gene was detected. In order to improve the antimicrobial activity, a sequential optimization strategy has been adopted. The antimicrobial potential was evaluated by following kinetics in a synthetic medium supplemented with different carbon and nitrogen sources. The highest bioactivity was obtained with corn steep liquor and sucrose. In a second step, Plackett-Burman experimental Design (PBD) was performed in order to determine the most significant culture parameters affecting the antimicrobial activity. It showed that corn steep liquor and KH2PO4 were the most influencing parameters. For maximum antimicrobial activity production, the response surface methodology (RSM) with Box-Behnken design (BBD) set the optimal values of the important variables at: 7 g / L of sucrose, 52 g / L of corn steep liquor and 0.9 g / L of KH2PO4, and an optimized medium was formulated. In these optimal conditions, an increase in antimicrobial activity production of 32% against methicillin resistant Staphylococcus aureus, 27% against Bacillus subtilis and 36% against Umbelopsis ramanniana was recorded. From a culture in batch fermenter using the optimized culture medium, the extraction of the antibiotic molecules was carried out with dichloromethane. The antimicrobial activity was detected in the aqueous phase unlike the reference work. Purification by HPLC with a hydrophilic column revealed the presence of seven active peaks. Among them, A1 and A5 with both antifungal and antibacterial activity, have been purified and partially characterized. The UV-visible spectra of the two molecules indicate the absence of polyenes. Proton nuclear magnetic resonance suggests that these 2 antibiotics have a glycosylated aromatic structure. The obtained results encourage the continuation of studies on antibiotics of the SA198 strain.La souche Saccharotrix tamanrassetensis DSM 45947, désignée SA198, est une nouvelle espèce d’actinobactérie d’origine saharienne, qui lors d’un travail précédent, s’est avérée produire dans le milieu complexe ISP2 trois nouvelles molécules antibiotiques à activité antifongique et antibactérienne. Notre travail a pour but d’explorer le potentiel antimicrobien de la souche, d’améliorer la production des molécules antimicrobiennes, de les purifier, et de les caractériser. La souche SA198 a montré une activité antagoniste intéressante, en particulier contre les agents pathogènes. La recherche de certains gènes de biosynthèse par PCR a permis de détecter la présence des gènes non-ribosomal peptide synthétases (NRPS). Pour améliorer la production de l’activité antimicrobienne, une stratégie d’optimisation séquentielle a été adoptée. Au départ, la production de cette activité (antifongique et antibactérienne) a été étudiée en suivant des cinétiques en milieu synthétique additionné de différentes sources de carbone et d’azote. L’activité la plus élevée a été obtenue avec le corn steep liquor et le saccharose. Par la suite, l’application du plan d’expérience de Plackett-Burman (PBD) a montré que le saccharose, le corn steep liquor et le KH2PO4 étaient les paramètres les plus importants influençant la production de l’activité antimicrobienne. Pour une production maximale de cette activité, la méthodologie de surface de réponse (RSM) avec le plan de Box-Behnken (BBD), a fixé les valeurs optimales des variables importantes à : 7 g/L de saccharose, 52 g/ L de corn steep liquor et 0,9 g/L de KH2PO4, et un milieu de culture optimisé est formulé. Dans ces conditions optimales, une augmentation de la production de l’activité antimicrobienne de 32% contre Staphylococcus aureus résistant à la méthicilline, 27% contre Bacillus subtilis et 36% contre Umbelopsis ramanniana, est observée. A partir d’une culture en fermenteur en mode discontinu, dans le milieu de culture optimisé, l’extraction des molécules antibiotiques est réalisée avec le dichlorométhane. L’activité antimicrobienne est détectée dans la phase aqueuse contrairement au travail de référence. La purification par HPLC avec une colonne hydrophile a révélé la présence de sept pics actifs. Deux d’entre eux, A1 et A5, à activité à la fois antifongique et antibactérienne ont été purifiés et caractérisés partiellement. Les spectres UV-visible des 2 molécules indiquent l’absence de polyènes. La résonance magnétique nucléaire du proton suggère que ces 2 antibiotiques possèdent une structure aromatique glycosylée. Les résultats obtenus encouragent la poursuite des études sur les antibiotiques de la souche SA198
