Two-dimensional gel electrophoresis of selenized yeast and autoradiography of Se-75-containing proteins

Two-dimensional high-resolution gel electrophoresis (2DE) has been applied to the fractionation of Se-75-containing proteins in yeast, grown in Se-75-containing medium, and autoradiography was used for detection of the Se-75-containing proteins. Gel filtration and ultrafiltration were used to check whether the selenium sidechains were stable in the presence of the chemicals used for lysis and 2DE. The mass distribution of the selenium-containing proteins was estimated by use of gel filtration and the results were compared with the distribution obtained by 2DE.
