Die Organisation der Blutgefäße im primären visuellen Kortex des Makaken und des Totenkopfäffchens und der Zusammenhang zur lokalen Stoffwechselaktivität

The quantitative and qualitative evaluation of the supplying blood vessels and the local capillary density in the visual cortex of macaques and squirrel monkeys, as well as the relationship of vascular density and oxidative metabolic capacity were the main topics of this thesis. Two publications showed locally different capillary densities and their correlation to the activity of the oxidative metabolism. The organization of the blood supply of the brain was investigated using a variety of different techniques. On the basis of corrosion casts, the morphology, the amount and the distribution of the supplying arteries and the draining veins was described. Additionally, an impression of the general organizational building principles of the vascular system was received. In histological brain sections the blood vessels were immunohistochemically labeled and visualized with a fluorescent dye. A nuclei stain on the same sections but in another color of the fluorescent spectrum enabled a reliable anatomical localization of the investigated regions. Another staining technique showed the local activity of cytochrome c oxidase, which is a mitochondrial enzyme and part of the respiratory chain reaction. In combination with the immunohistochemical staining of the blood vessels the tight correlation of metabolic activity and vascular density could be revealed. In general, the architecture of the cortical blood vessels of the monkey is very similar to that described for humans. The morphology of the large arteries and veins, their branching pattern to the capillary level and indications for blood flow regulating structures are equivalent amongst primates. Measurements of local vascular densities show an inhomogeneous distribution of the capillaries, with an increased density in the middle cortical layers and an overall higher blood vessel density in the primary visual area V1. It also turned out that the capillary density in particular correlates very well with the activity of the cytochrome oxidase, not only across cortical layers, but also in other subregions of the primary visual cortex with a noticeably increased cytochrome oxidase activity, namely the so-called "blobs". This suggests a potentially proportional relationship between oxidative metabolism and vascular density. It can be summarized that differences in vascular density of the cortex exist and that especially the primary sensory areas seem to be provided superiorly. This could have impact on the modern functional hemodynamic neuroimaging methods, whose signals stem from the blood stream and therefore underlie the restrictions of the anatomy of the system. The interpretation of such signals is often based upon the postulate of a homogenous perfusion, but particularly when comparing primary to non-primary sensory areas, this assumption could lead to false conclusions. The results presented in this thesis could contribute to achieve a more realistic evaluation of the measurable hemodynamic phenomena.Die quantitative und qualitative Beurteilung der versorgenden Blutgefäße und der lokalen Kapillardichten im visuellen Cortex von Makaken und Totenkopfäffchen, sowie der Zusammenhang von Blutgefäßdichte und oxydativer Stoffwechselkapazität waren Gegenstand der Untersuchungen, die in dieser kumulativen Dissertationsschrift dargelegt werden. In zwei Publikationen wurden der Nachweis lokal unterschiedlicher Kapillardichten und deren Korrelation zur sauerstoffbasierten Stoffwechselaktivität erbracht. Die Organisation der Blutversorgung im Gehirn wurde mit verschiedenen Techniken untersucht. Anhand von Ausgusspräparaten, die einen Negativabdruck des Blutgefäßsystems darstellen, konnten die Morphologie, die Anzahl und die Verteilung der zuführenden Arterien und der ableitenden Venen beschrieben werden, sowie ein Eindruck des generellen organisatorischen Aufbaus des Gefäßsystems gewonnen werden. In Hirnschnittpräparaten wurden die Blutgefäße immunhistochemisch markiert und mit einem fluoreszierenden Farbstoff visualisiert, eine ebenfalls fluoreszierende Kernfärbung derselben Präparate ermöglichte die zweifelsfreie hirnanatomische Verortung der untersuchten Gebiete. Eine weitere Färbetechnik zeigte die lokale Aktivität der Cytochrom c Oxidase, eines mitochondrialen Enzyms des oxydativen Stoffwechsels. Diese in Kombination mit der immunhistochemischen Blutgefäßfärbung durchgeführt ermöglichte die Korrelation von Stoffwechselaktivität und Gefäßdichte. Grundsätzlich ähnelt die Gefäßarchitektur im Cortex des Affen der beim Menschen beschriebenen. Die Morphologie der großen Arterien und Venen, das Verzweigungsmuster bis zur Kapillarebene, sowie Hinweise auf den Blutfluss regulierende Strukturen entsprechen denen anderer Primaten. Die Messung lokaler Gefäßdichten zeigte eine inhomogene Verteilung der Kapillaren, mit einer höheren Dichte in den mittleren kortikalen Schichten und einer insgesamt erhöhten Gefäßdichte im primären visuellen Areal V1. Es zeigte sich auch, dass speziell die Kapillardichte gut mit der Aktivität der Cytochromoxidase korreliert, nicht nur über die kortikalen Schichten hinweg, sondern auch in anderen, durch eine erhöhte Cytochromoxidase-Aktivität auffällige Bereiche im primären visuellen Cortex, den sogenannten "Blobs". Dies könnte auf einen möglicherweise proportionalen Zusammenhang zwischen oxydativer Stoffwechselaktivität und Blutgefäßdichte hindeuten. Insgesamt bleibt festzuhalten, dass Unterschiede in der Dichte der Blutgefäße im Cortex existieren, speziell die primären sensorischen Bereiche scheinen hier stärker versorgt zu sein. Dies könnte Einfluss auf die modernen, funktionellen hämodynamischen Bildgebungsverfahren haben, deren Signale aus der Blutbahn stammen und somit den Randbedingungen der Anatomie des Systems unterworfen sind. Bei der Interpretation solcher Messungen wird häufig vereinfacht von einer homogenen Durchblutung ausgegangen, doch vor allem bei Vergleichen zwischen primären und nicht-primären sensorischen Arealen könnte diese Annahme zu falschen Schlüssen führen. Die vorliegenden Ergebnisse könnten einer realistischeren Einschätzung der messbaren hämodynamischen Phänomene zuträglich sein
