Synthese von Benzomorphan Scaffolds durch Intramolekulare Buchwald-Hartwig Arylierung und ein Zugang zur Totalsynthese des Macrolids Queenslandon

First part represents a new strategy toward the important class of benzomorphans. The key bond formation is based on an intramolecular Buchwald-Hartwig enolate arylation reaction. Thus, alkylation of piperidones with ortho-bromobenzyl bromides provides the necessary substrates. In the presence of a palladium catalyst, a sterically hindered phosphane ligand, and a base, carbon-carbon bond formation to tricyclic benzomorphan derivatives takes place. After removal of the N-protecting group, derivatization reactions are possible. The second part of the dissertation contains the the preparation of the fully functionalized core structure of the antifungal macrolactone queenslandon using a novel strategy consisting of a glycolate aldol reaction and hydroboration of the corresponding enol ether followed by Suzuki cross-coupling with an iodostyrene. After conversion of the cross-coupling product to the seco acid, Mitsunobu macrolactonization and protecting group manipulations led to the queenslandon model compound.Der erste Teil der Arbeit beinhaltet eine neuartige Synthesestrategie für die bedeutende Klasse der Benzomorphane. Der Schlüsselschritt hierzu stellt eine intramolekulare Buchwald-Hartwig-Reaktion zur Arylierung von Enolaten dar. Die Alkylierung von Piperidonverbindungen mit ortho-Brombenzylbromiden führt zu den erforderlichen Ausgangssubstanzen. In Gegenwart eines Palladiumkatalysators sowie einem sterisch gehinderten Phosphanligand und einer Base findet die C-C-Knüpfung unter Bildung eines tricyclischen Benzomorphanderivates statt. Nach der Entfernung der N-Schutzgruppe können diese derivatisiert werden. Im zweiten Teil der Arbeit wird die Darstellung des antimykotisch wirkenden Macrolactons Queenslandon unter Verwendung einer neuartigen Synthesestrategie beschrieben. Diese beinhaltet eine Glykolat-Aldol-Reaktion sowie die Hydroborierung des entsprechnden Enol-Ethers gefolgt von einer Suzuki-Kupplung mit einem Iodstyren. Nach der Umsetzung des Produktes der Kreuzkupplung zu der Hydroxysäure führen Mitsunobu-Macrolactonisierung und Bearbeitung der Schutzgruppen zu der Modellverbindung Queenslandon
